ONCOTEST offers combination testing of substances in the monolayer assay.
Concept of drug combination testing:
- Analysis of viable cell number by PI assay
- Initial investigation of single drug application for IC50 determination
- Selection of combi layout 1,2 and/or 3) Combinations (A+B), as well as drug A and B alone are represented on one single microtiterplate
- Application of compounds by liquid handling robotic system

Three setups for combination analysis:
- A. Combination at fixed ratio
2 cytotoxic compounds at an equipotent ratio (e.g. at their IC50 values) at 10 concentrations Determination of Combination Index (CI) according Chou-Talalay by using CalcuSyn Software Evaluating synergism: CI<1 indicates synergism CI=1 indicates additive effect CI>1 indicates antagonism
- B. Combination at fixed concentration
cytotoxic compound A at 10 concentrations compound B at non / weak cytotoxic concentration(s) Evaluation by visual inspection of concentration-effect curve / shift of IC50/IC70 value
- C. Full combinatorial plate layout
2 cytotoxic compounds are tested in a matrix of 7 concentrations diagonals on a test plate represent equipotent ratio combinations coulums / rows represent fixed concentration combination Evaluation by (i) Combination Index according Chou-Talalay (ii) visual inspection of concentration-effect curves |
