Patient derived collection comprising 100 tumor cell lines and 120 xenografts growing subcutaneously and in part orthotopically in nude mice

Oncotest has an unique collection of more than 160 human tumors growing as subcutaneous xenografts in nude mice and additionally 300 nitrogen frozen human tumors.Tumors types comprise: Bladder, brain, colon, gastric, mammary, melanoma, ovary,pancreas, prostate, renal, sarcoma, lung (adeno, epidermoid, large cell and small cell), sarcomas (soft tissue and bone), uterus and hematological models.

Patient tumors were transplanted directly into nude mice and they are kept at low passages. In contrast to commercial cell lines and their derived tumors, the Oncotest tumor xenografts have maintained their typical tumor histology (with stroma, vasculature and central necrosis). Furthermore, their molecular characteristics mirror the original patient tumor and their drug sensitivity is highly predictive for the clinical situation.

Anticancer drug discovery is now focusing on target orientated approaches. Thus, target characterization of Oncotest’s tumors has been carried out for many selected tumor models and for many of the emerging targets in cancer therapy. Methods include ELISA, tissue and cell line arrays for IHC as well as gene expression profiling using Affymetrix chips. Targets comprise oncogenes (e.g. k-ras, c-myc), growth factors and receptors (e.g. EGF-R), drug resistance proteins (e.g. Mdr-1, GST), angiogenesis (e.g. VEGF), tumor-suppressor (e.g. p53, RB), metastasis related markers (e.g. uPA, cathepsin D, MMP).
